Reception:
rejection criteria:
- QNS “quantity not sufficient” (special condition for sodium citrate tubes to achieve the 1:9 ratio and avoid false results)
- Hemolyzed → plasma is red ish
- lipemic plasma (turbid)
- blood collected in a syringe
- urine collected in stool
- mismatch in label / unlabeled
- leakage
- form contamination from leakage
- plasma itself is contaminated
- correct tube
blood film:
- you want to achieve:
- tongue shape
- only 2/3rd of the slide smeared
- not thin, but also blood not touching the borders
Sickledex:
need:
- whole blood
- negative and positive control
- prepared reagent
procedure:
- add 2mL of reagent on each tube
- add 20 uL of (controls a+ sample0
- mix using droppers/pipettes
- wait for 5 minutes
principle of test is HbS solubility
positive test → presence of HbS which is insoluble in this case → turbid and lines not covered
